• MSCs-ATG5 therapy increases anti-inflammatory metabolite PGD2 and butyrate-producing bacteria, alleviating colitis symptoms.
• ATG5-engineered MSCs represent a promising strategy to improve MSC-based therapies for inflammatory bowel disease.
