• A mouse colon organoid model of ulcerative colitis was successfully established using lipopolysaccharide induction.
• Lipopolysaccharide at concentrations of 225-275 μg/mL significantly increased pro-inflammatory cytokines (IL-6, TNF-α) and decreased tight junction protein expression.
• The organoid model recapitulates key features of ulcerative colitis, including epithelial barrier disruption and inflammatory response.
• This model provides a valuable platform for drug screening and mechanistic studies of ulcerative colitis.
